Human Gene TNFRSF10A (ENST00000221132.8) from GENCODE V44
  Description: Homo sapiens TNF receptor superfamily member 10a (TNFRSF10A), mRNA. (from RefSeq NM_003844)
RefSeq Summary (NM_003844): The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor is activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL), and thus transduces cell death signal and induces cell apoptosis. Studies with FADD-deficient mice suggested that FADD, a death domain containing adaptor protein, is required for the apoptosis mediated by this protein. [provided by RefSeq, Jul 2008].

  ID: TR10A_HUMAN
DESCRIPTION: RecName: Full=Tumor necrosis factor receptor superfamily member 10A; AltName: Full=Death receptor 4; AltName: Full=TNF-related apoptosis-inducing ligand receptor 1; Short=TRAIL receptor 1; Short=TRAIL-R1; AltName: CD_antigen=CD261; Flags: Precursor;
FUNCTION: Receptor for the cytotoxic ligand TNFSF10/TRAIL. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. Promotes the activation of NF- kappa-B.
SUBUNIT: Can interact with TRADD and RIPK1. Interacts with ARAP1.
INTERACTION: Q96P48:ARAP1; NbExp=4; IntAct=EBI-518861, EBI-710003; Q14790:CASP8; NbExp=9; IntAct=EBI-518861, EBI-78060; P50591:TNFSF10; NbExp=8; IntAct=EBI-518861, EBI-495373;
SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.
TISSUE SPECIFICITY: Widely expressed. High levels are found in spleen, peripheral blood leukocytes, small intestine and thymus, but also in K-562 erythroleukemia cells, MCF-7 breast carcinoma cells and activated T-cells.
SIMILARITY: Contains 1 death domain.
SIMILARITY: Contains 3 TNFR-Cys repeats.

-  Orthologous Genes in Other Species
  Orthologies between human, mouse, and rat are computed by taking the best BLASTP hit, and filtering out non-syntenic hits. For more distant species reciprocal-best BLASTP hits are used. Note that the absence of an ortholog in the table below may reflect incomplete annotations in the other species rather than a true absence of the orthologous gene.
