Evidence: Stimulation of endothelial cells by FGF2 causes also the shedding of cell surface membrane vesicles containing MMP-2 and MMP-9 together with the two MMP inhibitors TIMP-1 and TIMP-2. These vesicles stimulate capillary-like structure formation when added to endothelial cells seeded on a solubilized basement membrane preparation (Matrigel) [26].

Evidence: FGF2 [50] and FGF8b [17] can enhance endothelial cell reorganization onMatrigel. The process requires the activation of the proteolytic machinery, including type IV collagenase(s) and TIMPs production [51] as well as uPA and PAI-1 upregulation [52], a6b1 integrin receptor engagement [53], PECAM-1 [54], and activation of the small GTPase Rac [55].

:
Furthermore, in both cell types, the secretion of
TIMP-2 was
enhanced by tPA and PDGF, but not by uPA or
bFGF
Toschi et al., Mol Biol Cell 2001
(Acquired Immunodeficiency Syndrome...) :
These effects are due to the activation of the membrane-type-1-matrix-metalloproteinase and to the
induction of the membrane bound
tissue inhibitor of metalloproteinase-2 ( TIMP-2 ) by Tat and
bFGF combined, but also to Tat mediated inhibition of both basal or bFGF induced TIMP-1 and -2 secretion
Wen et al., Life Sci 2002
:
Northern blot and nuclear run-off assay showed that
bFGF not only
stimulated the expression of migration related matrix metalloproteinase-2 (MMP-2) and
tissue inhibitor of metalloproteinase-2 ( TIMP-2 ) but also enhanced the transcription of proliferation related osteopontin (OPN) and c-jun genes in the quiescent VSMC
Asanuma et al., Kidney Int 2002
:
In addition,
bFGF enhanced the secretion of
TIMP-2 ( 2.75-fold at 20 ng/mL ) and TGF-beta1 enhanced the secretion of cystatin C ( 2.32-fold at 20 ng/mL )
Yasui et al., Digestion 2004
:
FGF-2 stimulated MMP-1, MMP-3 and TIMP-1 secretion, but did not
affect MMP-2 or
TIMP-2 secretion
Nakoman et al., Biochimie 2005
(Carcinoma...) :
The
effect of
bFGF on MMP-2,
TIMP-2 , total collagen, and type I collagen levels of normal and carcinoma lung fibroblast cultures was investigated at 0, 10, and 100 ng/ml. Statistical analysis was carried out using the Mann-Whitney-U test and possible correlations were searched using the Spearman correlation analysis method ... It may be concluded that
bFGF does not
affect MMP-2,
TIMP-2 , total collagen, and type-1 collagen levels in fibroblast cultures grown from human carcinoma and normal lung tissues
